RESUMEN

Increasingly hyperprolific sows and the need to reduce antibiotics represent challenges in pig farming. The aim of this work was to determine the effects of a postbiotic obtained from inactivated and stabilized whole-cells of Saccharomyces cerevisiae, administered during the sow's gestation, on the performance of the mother and litter. Maternal feed intake, productive parameters, colostrum quality and post-weaning piglets' health were assessed, including antibiotic consumption. The trial involved 183 sows, divided into two groups: (1) sows fed with a daily supplementation of postbiotic during gestation (n = 90); (2) sows without any supplement (n = 93). Piglets were followed up at two different post-weaning sites. The lactation efficiency of the treated sows improved by +5.9% (41.3 ± 11.4 vs. 35.4 ± 11.6%; p = 0.011). Lactating piglets' mortality was lower in the treated group (25.1 ± 16.7 vs. 28.8 ± 14.4%; p = 0.048). The same tendency was shown in both the weaning sites, together with a reduced antibiotic consumption in weaning site 1 (0.72 ± 0.25 vs. 1.22 ± 0.30 DDDvet/PCU; p = 0.047). The results suggest the role of this postbiotic administered to the mother in improving the health status of the piglets. Furthermore, lactation efficiency is suggested as an interesting parameter for assessing the efficiency of farming.

In a previous study, a semi-quantitative risk assessment was developed to rank pig holdings in terms of likelihood of introducing African swine fever virus (ASFV) by assessing their compliance with biosecurity and exposure to geographical risk factors. The method was initially developed for confined pig holdings, but given that ASF is endemic in wild boar of several countries, we modified the approach to make it suitable for free-range farms as well. In the current study, a total of 41 outdoor pig farms were assessed in an area where exposure to wild boar was generally high (density from 2.3 to 10.3 wild boar per Km2). As expected, non-compliance with biosecurity measures was frequent in outdoor farms, and the frequency of non-compliance indicated that the absence of adequate separation of pigs from the external environment was the major weakness in the farms assessed. In 46.3% of them, there was no fence or, if present, it was not adequate to avoid contact with wild boar. However, the approach adopted proved to be suitable for identifying intervention priorities to mitigate the risk of ASFV spread in free-range pig herds and for identifying the weaknesses of individual farms, as recommended by EFSA in 2021, which suggests implementing tools to improve biosecurity by favoring higher-risk farms.

In livestock, the importance of hygiene management is gaining importance within the context of biosecurity. The aim of this study was to monitor the implementation of biosecurity and hygiene procedures in 20 swine herds over a 12-month period, as driven by tailor-made plans, including training on-farm. The measure of adenosine triphosphate (ATP) environmental contents was used as an output biomarker. The presence of livestock-associated methicillin-resistant Staphylococcus aureus (LA-MRSA) and extended-spectrum ß-lactamase producing Escherichia coli (ESBL-E. coli) was also investigated as sentinels of antibiotic resistance. A significant biosecurity improvement (p = 0.006) and a reduction in the ATP content in the sanitised environment (p = 0.039) were observed. A cluster including 6/20 farms greatly improved both biosecurity and ATP contents, while the remaining 14/20 farms ameliorated them only slightly. Even if the ESBL-E. coli prevalence (30.0%) after the hygiene procedures significantly decreased, the prevalence of LA-MRSA (22.5%) was unaffected. Despite the promising results supporting the adoption of tailor-made biosecurity plans and the measure of environmental ATP as an output biomarker, the high LA-MRSA prevalence still detected at the end of the study underlines the importance of improving even more biosecurity and farm hygiene in a one-health approach aimed to preserve also the pig workers health.

African swine fever (ASF) is a severe haemorrhagic infectious disease affecting suids, thus representing a great economic concern. Considering the importance of the early diagnosis, rapid point of care testing (POCT) for ASF is highly demanded. In this work, we developed two strategies for the rapid onsite diagnosis of ASF, based on Lateral Flow Immunoassay (LFIA) and Recombinase Polymerase Amplification (RPA) techniques. The LFIA was a sandwich-type immunoassay exploiting a monoclonal antibody directed towards the p30 protein of the virus (Mab). The Mab was anchored onto the LFIA membrane to capture the ASFV and was also labelled with gold nanoparticles for staining the antibody-p30 complex. However, the use of the same antibody for capturing and as detector ligand showed a significant competitive effect for antigen binding, so required an experimental design to minimize reciprocal interference and maximize the response. The RPA assay, employing primers to the capsid protein p72 gene and an exonuclease III probe, was performed at 39 °C. The limit of detection of the method was assessed using a plasmid encoding the target gene and resulted in 5 copy/µL. The new LFIA and RPA were applied for ASFV detection in the animal tissues usually analysed by conventional assays (i.e., real-time PCR), such as kidney, spleen, and lymph nodes. A simple and universal virus extraction protocol was applied for sample preparation, followed by DNA extraction and purification for the RPA. The LFIA only required the addition of 3% H2O2 to limit matrix interference and prevent false positive results. The two rapid methods (25 min and 15 min were needed to complete the analysis for RPA and LFIA, respectively) showed high diagnostic specificity (100%) and sensitivity (93% and 87% for LFIA and RPA, respectively) for samples with high viral load (Ct < 27). False negative results were observed for samples with low viral load (Ct > 28) and/or also containing specific antibodies to ASFV, which decreased antigen availability and were indicative of a chronic, poorly transmissible infection. The simple and rapid sample preparation and the diagnostic performance of the LFIA suggested its large practical applicability for POC diagnosis of ASF.

A semi-quantitative risk assessment was developed to classify pig farms in terms of the probability of introduction of African swine fever virus (ASFV). Following on-farm data collection via a specific checklist, we applied a modified failure mode and effect analysis (FMEA) to calculate the risk priority codes (RPC's), indicating increasing risk levels ranging from 1 to 5. The importance of biosecurity measures was attributed by experts. To consider geographic risk factors, we classified pig farms based on local density of farmed pigs, and on the estimated wild boar population density. The combination of RPC's with geographical risk factors resulted into a final ranking of pig farms in terms of the risk of ASFV introduction. Furthermore, the estimation of frequency and levels of non-compliance with biosecurity measures was used to identify weak points in risk prevention at farm level. The outcome of the risk assessment was affected by choices in assigning non-compliance scores and importance to specific components of biosecurity. The method was applied in 60 commercial farms in major pig production areas in Italy. Furthermore, we applied a reduced version of our checklist in 12 non-commercial/small commercial (≤20 pigs) farms in the northern Apennines. In commercial farms, highest RPC's were obtained for biosecurity measures associated with personnel practices and farm buildings/planimetry. Intervention should be addressed to training of personnel on biosecurity and ASF, to avoid contacts with other pig herds, and to improve practices in the entrance into the farm. Sharing trucks with other farms, and loading/unloading of pigs were other weak points. Fencing was classified as insufficient in 70% of the commercial farms. Among these farms, breeding units were characterised by the lowest risk of ASFV introduction (although differences among median ranks were not statistically significant: P-value = 0.07; Kruskal-Wallis test), and increasing herd size was not significantly correlated with a higher risk (Kendall's τ = -0.13; P-value = 0.14). Density of farmed pig was greatest in the main pig production area in northern Italy. Conversely, exposure to wild boars was greatest for non-commercial/small commercial farms on the Apennines, which were also characterised by non-compliance with critical biosecurity measures.

Tail biting is an economical and behavioral problem in the pork production system worldwide and systematic tail docking has been applied for decades to decrease the risk of its onset. However, legal and market-driven requirements are leading pig producers to rear undocked animals. The aim of this work was to monitor tail, pluck (lungs, pleurae, and liver), stomach, carcass, and thigh lesions in slaughtering pigs belonging to either docked or undocked batches. A total of 525 batches were evaluated at slaughter: 442 docked and 83 undocked batches. The presence of tail lesions was only recorded in undocked batches (44.0 ± 0.402 vs. 0.2 ± 0.2% compared to docked ones, p < 0.001), with a prevalence of severe chronic lesions of 27.3% ± 0.032, suggesting that more and alternative wide efforts to manage long-tailed animals are needed. On the contrary, docked animals showed more frequent ear lesions (9.6% ± 0.037 vs. 4.6% ± 0.019; p = 0.0001). Severe lung lesions were found more frequently in undocked animals (9.2% ± 0.043 vs. 6.6% ± 0.011, p = 0.006), as well as gastric ulcers (26.1% ± 0.021 vs. 20.3% ± 0.37, p = 0.006). These lesions might share the same predisposing factors of tail lesions; the latter might be investigated as an iceberg indicator for other pathological conditions in undocked pigs and eventual causal association among lesions in these organs should be explored.

Limited data are available regarding animal-based biomarkers over time as outcomes of biosecurity in pig farms. The aim of this study was to gain an insight into the biosecurity implementation in a convenience sample of 15 swine herds, and to describe potential biomarkers of interest; inputs from a systematic evaluation of biosecurity implementation were used to develop tailor-made biosecurity protocols monitored over a 12-month period. The farms' implementation was then described, and animal-based biomarkers were explored as output parameters. A significative biosecurity improvement was observed at the end of the study (p = 0.047), in particular in the professional zone (p = 0.012). Four clusters of farms were identified for their progress on biosecurity implementation by means of Principal Component Analysis (PCA) and Hierarchical Cluster Analysis (HCA): 4/15 farms improved their biosecurity only in the professional zone, 8/15 showed scarce/null improvement of total biosecurity, 2/15 worsened their biosecurity, and 1/15 greatly improved biosecurity. The farm biosecurity profiles showing an improvement included farms with a reduction in lung lesions and scars at slaughter at the end of the study. The results suggest that a systematic evaluation of biosecurity is a useful approach to formulate tailor-made biosecurity plans and monitor their implementation; biomarkers might bring insight into the outcomes of biosecurity.

Oedema disease (ED) caused by Shiga-toxin-producing E. coli in pigs is a serious life-threatening disease, particularly among weaned piglets. When a preventive protocol is adopted in a specific farm, interpretation of effectiveness is often complicated in field conditions due to natural or "common cause" variation. For this reason, in this study a Statistical process control (SPC) approach was used to retrospectively evaluate the application of an ED preventive protocol (lower protein diet, ad-libitum fiber, vaccination at 5 days of age) in an infected commercial piglets' weaning site. The analysis was established over a 9-years period (n = 75 consecutive batches; 1,800 weaners per batch) using mortality for each batch as the key parameter of health and production; the statistics and the control limits (mean ± 3-fold sd; UCL, upper control limit; LCL, lower control limit) were based on data from the first 28 batches (Period 1) before the onset of the first ED clinical signs. The charts allowed the detection of defined out of control batches (i.e., with mortality out of the intervention limits) from batch 29 ongoing, exploring a Period 2 (unstable production and ED clinical signs; 36 batches) and a Period 3 (application of the ED preventive protocol; 11 batches). Mortality evaluation using SPC revealed a production system defined under-control (mean moving range bar = 1,34%; UCL = 4,37%; LCL = 0%) during Period 1. During Period 2, charts lost the state of statistical control, as showed by several signals of special cause variation due to the ED outbreak. Period 3 was characterized again by a state of statistical control, where no signals of special cause variation was showed. In conclusion, the retrospective application of SPC charts in the present study was able to confirm the efficacy of an ED preventive protocol in reducing mortality in a piglets' weaning site. SPC charting is suggested as an useful tool to provide insights into relationships between health, managerial, and welfare decision and some selected iceberg parameters in livestock.

Piglet castration results in acute pain and stress to the animal. There is a critical need for effective on-farm methods of pain mitigation. Local anaesthesia using Tri-Solfen® (Animal Ethics Pty Ltd., Melbourne, Australia), a topical local anaesthetic and antiseptic formulation instilled to the wound during surgery, is a newly evolving on-farm method to mitigate castration pain. To investigate the efficacy of Tri-Solfen®, instilled to the wound during the procedure, to alleviate subsequent castration-related pain in neonatal piglets, we performed a large, negatively controlled, randomised field trial in two commercial pig farms in Europe. Piglets (173) were enrolled and randomised to undergo castration with or without Tri-Solfen®, instilled to the wound immediately following skin incision. A 30 s wait period was then observed prior to completing castration. Efficacy was investigated by measuring pain-induced motor and vocal responses during the subsequent procedure and post-operative pain-related behaviour in treated versus untreated piglets. There was a significant reduction in nociceptive motor and vocal response during castration and in the post-operative pain-related behaviour response in Tri-Solfen®-treated compared to untreated piglets, in the first 30 min following castration. Although not addressing pain of skin incision, Tri-Solfen® is effective to mitigate subsequent acute castration-related pain in piglets under commercial production conditions.

The analysis of antimicrobial use (AMU) data in livestock allows for the identification of risk factors for AMU, thereby favoring the application of responsible AMU policies on-farm. Herewith, AMU in 36 finishing pig farms in northern Italy from 2015-2017 was expressed as defined daily doses for Italian pigs (DDDita) per population correction unit (DDDita/100kg). A retrospective analysis was then conducted to determine the effects of several husbandry practices on AMU. Overall, AMU ranged between 12 DDDita/100kg in 2015 and 8 DDDita/100kg in 2017, showing no significant trends, due to the large variability in AMU between farms. However, a 66% AMU reduction was observed in 19 farms during 2015-2017. Farm size, number of farm workers, air quality, average pig mortality, and presence of undocked pigs on the farm had no significant effects on AMU. Rather, welfare-friendly farms had 38% lower AMU levels (p < 0.05). In conclusion, animal welfare management seems to be relatively more important than farm structure and other managerial characteristics as drivers of AMU in finishing pig farms.

The aim of this study was to analyze the feasibility and reliability of using hair as a matrix to determine the dehydroepiandrosterone (DHEA) and sexual steroid concentrations and the cortisol/DHEA ratio in fattening pigs. The results could be also used to plan future research to identify threshold values in order to set up strategies to control the allostatic load and increase the resilience of fattening pigs before slaughter. The study was conducted on 107 commercial crossbred rearing pigs. The hair samples were taken by shaving at the age of 36 weeks, and concentrations of the hormones were measured using a solid-phase microtiter radioimmunoassay. Females had significantly higher cortisol levels (p < 0.01), significantly lower DHEA concentrations (p < 0.05) and significantly higher cortisol/DHEA ratios (p < 0.01) than barrows. Progesterone was significantly higher in gilts than in barrows (p < 0.01). Testosterone and 17ß-estradiol were significantly higher in barrows than in gilts (p < 0.05). If future research can produce threshold values for the different markers examined, the evaluation of animals under subclinical stress conditions will be possible.

In order to evaluate at the slaughterhouse external carcass lesions on heavy pigs (170 kg) as potential welfare indicators, and the prevalence of ham defects determining ham exclusion from Protected Designation of Origin (PDO) markets, 732 pig batches from northern Italy were monitored during a 12-month period, and then processed analysing the effect of slaughter season, overnight lairage, and production type. On the slaughter line, skin scratches were separately scored in the posterior region (defined as the area including the hind legs and the tail) and the anterior one (as the remaining area), while the whole carcass was examined for external hematomas. Chronic ear and tail lesions referable to the rearing phase, and bursitis were recorded as retrospective welfare indicators. The annual median prevalence of carcasses per batch with severe anterior scratches was 64% while 46.4% had severe posterior scratches. The highest autumn score for both skin scratches (P < 0.001) and traumatic ham defects (P = 0.005) is reflected in the positive correlation between severe posterior scratches and ham hematomas (r2 = 0.27; P < 0.001). Overnight lairage batches resulted in higher prevalence for scratches, while among ham defects only veining increased. Among binary records, only ear lesions were frequently recorded (annual median = 10%). A comparison analysis between pigs in and out of PDO circuit was performed to evaluate the variation related to the different genetics, showing differences for ear and tail lesions and for almost all the considered ham defects. The present study confirms that skin lesions represent a problem also for heavy pigs and that overnight lairage and season can affect their prevalence, with the associated possibility to give ham defects. Ear lesions are suitable to be used as retrospective welfare indicator, while tail lesions usage is nowadays limited by the extensive use of tail docking.

Tail biting in pigs has been an identified behavioural, welfare and economic problem for decades, and requires appropriate but sometimes difficult on-farm interventions. The aim of the paper is to introduce the Classification and Regression Tree (CRT) methodologies to develop a tool for prevention of acute tail biting lesions in pigs on-farm. A sample of 60 commercial farms rearing heavy pigs were involved; an on-farm visit and an interview with the farmer collected data on general management, herd health, disease prevention, climate control, feeding and production traits. Results suggest a value for the CRT analysis in managing the risk factors behind tail biting on a farm-specific level, showing 86.7% sensitivity for the Classification Tree and a correlation of 0.7 between observed and predicted prevalence of tail biting obtained with the Regression Tree. CRT analysis showed five main variables (stocking density, ammonia levels, number of pigs per stockman, type of floor and timeliness in feed supply) as critical predictors of acute tail biting lesions, which demonstrate different importance in different farms subgroups. The model might have reliable and practical applications for the support and implementation of tail biting prevention interventions, especially in case of subgroups of pigs with higher risk, helping farmers and veterinarians to assess the risk in their own farm and to manage their predisposing variables in order to reduce acute tail biting lesions.

Abattoir post-mortem inspections offer a useful tool for the development and monitoring of animal health plans and a source of data for epidemiological investigation. The aim of the present work was to develop an abattoir benchmarking system which provides feedback on the prevalence and severity of lesions of the pluck (lung, pleura and liver) in batches of pigs to inform individual producers and their veterinarians of the occurrence of pathological conditions affecting their herds. The weekly collection of data throughout a year (from September 2014 to September 2015) supported the further aim of providing benchmark values for the prevalence of lesions and their seasonality in Italian heavy pig production. Finally, correlations and redundancies among different lesions were evaluated. In total, 727 batches of heavy pigs (around 165kg live weight and 9 months of age) derived from 272 intensive commercial farms located in Northern Italy were monitored. Within each batch, an average number of 100 plucks was individually scored, assigning a value for lesions of lungs (0-24), pleura (0-4) and liver (1-3). Presence of lung scars, abscesses, consolidations, lobular/chessboard pattern lesions and pleural sequestra was also recorded. Statistical analysis showed a strong farm effect (36-68% of variation depending of the lesion) and a seasonal effect on all lesions. Winter showed the lowest percentage of severe lung and pleural lesions (P<0.001 and P=0.005), whereas lung scars from older lesions (P=0.003), as well as severe hepatic lesions (P<0.001), were reduced in autumn. In order to allow effective benchmarking of each farm in a determined health class, scores for each quartile of the population are reported. Whilst such a benchmarking scheme provides useful data for herd health management, challenges of repeatability of scoring and cost of implementation need to be overcome.

Many piglets are exposed to potentially painful husbandry procedures within the first week of life, including tail docking and castration, without the provision of either anesthesia or analgesia. The assessment methods used to evaluate pain experienced by piglets are often affected by low specificity and practical limitations, prompting the investigation of alternative methodologies. The assessment of changes in facial expression following a painful event has been successfully applied to several species. The objective of this pilot study was to evaluate the utility of a Grimace Scale applied to neonatal pigs to evaluate pain evoked by tail docking and castration. Eight female piglets, Sus scrofa domesticus (Landrace/Large White X synthetic sire line) underwent tail docking and 15 male piglets (75% Large White and 25% Belgian Landrace) were exposed to the castration procedure. Clear images of the faces of the piglets were collected immediately pre- and post-procedure. The images were used by experienced observers to identify facial action units (FAUs) which changed in individuals over this period, and a scoring scale was depicted in a training manual. A set of randomly selected images were then combined in a scorebook, which was evaluated after training by 30 scorers, blind to the treatment. The scale for most FAU was used with a high level of consistency across all observers. Tail docking induced a significant change (P < 0.05) in free moving piglets only in the "orbital tightening" FAU, whereas no change in any unit was observed in castrated piglets, which were restrained at the time of assessment. In this initial stage of development, orbital tightening seems to have the potential to be applied to investigate painful conditions in neonatal pigs. Nonetheless, more studies are needed to assess its full effectiveness and to evaluate the influence of possible confounds (e.g., handling stress) on the observed changes in FAUs.

Adopting a 2 × 2 × 2 factorial design, this study evaluated whether continuous straw provision by racks, tail docking and gender (barrows vs. females) have an effect on the prevalence of lung lesions and oesophago-gastric ulcer (OGU) visually scored at slaughter in 635 Italian heavy pigs (169 ± 4 kg). The lung lesions were very low (72% of pigs with score 0), and were not significantly different among the experimental groups. Overall, OGU was diagnosed in 47% of the pigs. The consumption of small amounts of straw (70 g/day/pig) represented a protective factor against the onset of OGU (OR: 0.27). Barrows were more likely than females to have OGU (OR: 1.52), while no significant differences between docked and undocked pigs were detected. Nevertheless, the presence of straw acted as a protective factor particularly in undocked pigs (OR: 0.16), suggesting that in this group the absence of rooting material may have a stronger effect on welfare.

RESUMEN

This study evaluated whether the specific heavy pig rearing context allowed the fattening of undocked pigs without an outbreak of tail biting. At the same time, gender and straw availability (small amounts) were considered to understand their possible interactions with tail presence in the display of tail biting. A 2 × 2 × 2 factorial design was adopted to test the effects of these factors on blood parameters, behaviour and tail/ear lesions. Few interactions among factors were detected. Undocked pigs showed lower cortisol (P<0.02), lying behaviour (P<0.001), and higher risk of tail/ear biting (weeks 3 and 9), but lower risk of tail lesions (week 14). Straw increased the motivation for exploring (P<0.001), reduced serum haptoglobin (P<0.001) and the risk for tail biting (weeks 3, 9, 18) and ear biting (weeks 3, 9). Results highlight the importance of straw as an environmental enrichment and seem to indicate that fattening undocked heavy pigs is possible.
